Preparation of L-PLA Microspheres containg 5-FU using Supercritical Carbon Dioxide. |

5-fluorouracil (5-FU) is widely used chemotherapeutic agent for the treatment of metastatic breast cancer. To obtain maximal efficacy with minimal systemic side-effects, many studies have been carried out to achieve the sustained release of 5-FU. Thus, a water-soluble anti-cancer drug, 5-FU was encapsulated in microspheres of poly(L-lactide) using supercritical fluid. Micro- or nano-size drug particles were produced using aerosol solvent extraction system (ASES) and determine the physicochemical characteristics of the developed microspheres. The operating conditions were 35°c of temperature, 130 bar of pressure and 5~40% (w/w) of drug/polymer concentrations in solutions. Our results in dicate that ASES process was more efficient than traditional method. |
