We designed transparent, stable and insoluble silk fibroin films by blending with glycerol as an alternative scaffold for bioengineering cornea. The morphological and structural properties of films were analyzed. The surface cracks in silk fibroin films with glycerol were observed by microscope. Glycerol content affected in transparency of each glycerol/silk fibroin films (G/SF), but it was not significant. In vitro biological compatibility was studied using CEnCs. Even though differences occurred according to the ratio of glycerol and silk fibroin, they did not show any significant differences on cell morphology and well-expressed their functional proteins which are regulated functions of CEnC. Overall, the results suggest that silk fibroin offers good environment on growth of CEnCs and glycerol plays a role to improve physical properties. Overall results showed that G/SF can be used as a suitable alternative for high quality corneal tissue expansion and transplantation. |
