제목 Galactosylated Polyethylenimine -graft-poly(vinyl pyrrolidone) as a Hepatocyte-Targeting Gene Carrier
초록 Non-viral gene carriers in gene therapy such as cationic polymers may overcome the current problems associated with viral gene carrier in safety, immunogenicity and mutagenesis. In this study, polyethylenimine (PEI) was used as a gene carrier and conjugated with lactose bearing galactose group for hepatocyte targeting and grafted with poly(vinyl pyrrolidone) (PVP) as a hydrophilic group to reduce cytotoxicity of PEI and prevent aggregation of complex from proteins in vivo. The galactosylated PEI-PVP (Gal-PEI-PVP)/DNA complex was confirmed using agarose gel electrophoresis and DNase I experiment. The sizes and zeta potentials of the complex were measured by dynamic light scattering. The cytotoxicity of Gal-PEI-PVP in HepG2 cells was evaluated. The Gal-PEI-PVP/DNA complex was transfected into HepG2 cells having asialoglycoprotein receptors (ASGRs), indication of specific interaction between ASGRs on HepG 2 cells and galactose ligands on Gal-PEI-PVP.
