Preparation of Alginate/Xyloglucan Beads for Bioartificial Liver |

The carbohydrate polymer alginate is known as one of the biocompatible extracellular matrices (ECM). Alginates form hydrogels at room temperature and neutral pH, due to the G-block in alginate rapidly crossed by a divalent cation. Therefore alginate was also used as a three dimensional ECM for immobilization of cells in tissue engineering. However, hepatocytes encapsulated in alginate are of low viability and do not maintain the long term liver functions due to the lack of cell-matrix and cell-cell interactions within alginate. Xyloglucan having galactose ligands is expected to interact with asialoglycoprotein receptors of hepatocytes. When xyloglucan polysaccharide derived from tamarind seed is partially degraded by β-galactosidase, the resultant product exhibits thermally reversible gelation in dilute aqueous solution. Hepatocytes in alginate/xyloglucan (AL/XG) beads will be able to produce more cell-specific functions than alginate beads alone. In this study, AL/XG beads were prepared and characterized. Also, hepatocytes were entrapped in AL/XG beads. Furthermore, cell viability and cell function of the entrapped hepatocytes were checked. |
