Branched polyethylenimine (bPEI) has many amine group. it used high gene transfection agent. However, bPEI utilization as gene carrier was limited because of high cytotoxicity. To overcome these, chitosan was introduced to bPEI by carbonyldiimidazol as coupling agent. The chemical structure of chitosan-g-bPEI analyzed by 1H-NMR. The binary and ternary complexes particle size, zeta potential and mophological property was confirmed by DLS, zeta sizer and TEM. Moreover, transfection efficiency and cellular uptake showed that ternary complexes more than binary complexes. The targeting effect of ternary complexes was confirmed by competition assay in CD44-receptor overexpressed HCT116 cell line. These results suggest that ternary complexes is superb gene carrier with targeting effect CD44-receptor positive cancer cell line. (No. NRF-2016H1D5A1910499). |
