Bioinspired pH- and Temperature-Responsive Injectable Adhesive Hydrogels with Polyplexes Promotes Skin Wound Healing |

We developed a bioinspired pH- and temperature-responsive in situ-forming injectable hydrogel depot that could control the delivery of DNA-bearing polyplexes for versatile biomedical applications. The pH- and temperature-responsive copolymer, composed of hydrophilic poly(ethylene glycol) (PEG) and pH- and temperature-responsive poly(sulfamethazine ester urethane) (PSMEU), has been synthesized as in situ-forming injectable hydrogel. PEG-PSMEU copolymers exist as low viscous sols at high pH and room temperature (pH 8.5, 23 oC); whereas, they transform to a viscoelastic gel at the physiological condition (pH 7.4, 37 oC). Owing to the presence of urethane linkages, the PEG-PSMEU possesses excellent adhesion strength to wide range of surfaces including glass, plastic and fresh organs. The bioinspired pH- and temperature-responsive in situ-forming injectable hydrogel may be promising for biomedical applications such as controlled delivery vehicle, tissue adhesive and tissue regeneration. |
