Piroxicam(PX) is one of nonsteroidal anti-inflammatory drugs (NSAIDs) and has been used for musculoskeletal disorders, dysmenorrhea, and postoperative pain. PX is usually treated by oral administration, but there were many adverse effects such as gastrointestinal ulceration, bleeding perforation, and renal disorder. Herein, we designed a thermo-responsive hydrogel for intra-articular injection which exhibits a sustained drug release. The hydrogel is composed of thermosensitive polymer (TSP) and biocompatible polymer (BP) by simple mixing in aqueous solution. In this study, we demonstrated that a novel NSAID delivery formulation for PX-incorporated BP and TSP hydrogel (BPT) not only exhibits an excellent mechanical-strength, but also results sustained release PX. Therefore, we believed that the BPT hydrogel could be a successful PX carrier for the various disease treatments. |
