제목 Redox-sensitive and stabilized heparin-Pluronic nanogels for efficient intracellular drug delivery
초록 Polymeric nanocarriers for intracellular delivery have many limitations, such as short half-life, non-specific drug release, although it has been continuously potentiated. Our group also has developed a potential nanocarrier, nano-sized heparin-based hydrogels, suffering from low stability and non-specific release of drugs. Herein, thus a heparin-Pluronic (HP) nanogel was strengthened by crosslinking with disulfide bond. The disulfide crosslink in HP nanogel may enable glutathione-sensitive release of a drug in the intracellular region and long-lasting circulation without dissociation. In vitro release study showed that the drug release time extended about 3 folds comparing to unmodified heparin-Pluronic nanogels. Besides, the release time increased 2.5 folds in presence of 5.0 mM glutathione than the absence of glutathione. These results suggest that disulfide-crosslinking may facilitate the improved stability and release profile of HP nanogels for efficient intracellular delivery.
