The Polymerization of Fibrinogen Coupled with MWNTs using plasma transglutaminase (Factor XIII) |

Carbon nanotubes (CNTs) have many interesting and unique properties to be used in a variety of biological and biomedical systems. For the development of its potential applications, various methods of functionalization of CNTs have being developed to interface CNTs with organic and inorganic materials, as well as biological macromolecules. The latter is gaining popularity because of potential applications in biosensor development, bioelectrochemistry, biomedical devices, and cellular delivery of peptides and proteins. In this study, we conducted the amide coupling between MWNT-COOH and fibrinogen (FBG). FBG is an acute phase defensive protein that participates as a core protein in the final phase of blood coagulation. The main function of FBG is to promote the formation of three-dimensional network of fibrin fibers that forms blood clots. After coupling of FBG with MWNT-COOH, the polymerization of MWNT-FBG to MWNT-Fibrin was conducted successfully adding activated plasma transglutaminase (Factor XIII). MWNT-FBG and MWNT-Fibrin were characterized by using FT-IR spectroscopy, transmission electron microscope (TEM), and energy dispersion of X-ray spectroscopy (EDX) coupled with transmission electron microscope(TEM). In conclusion, the MWNT-FBG still maintained the fibrinogen property, where Factor XIII were able to polymerized the MWNT-FBG to MWNT-Fibrin. |
