제목 Formulations Based on Hyaluronic Acid and TNF-Related Apoptosis-Inducing Ligand as the Injectable Protein Delivery System
초록 Although Tumor Necrosis Factor-Related Apoptosis-Inducing Ligand (TRAIL) is considered as the potent therapeutics for tumors, its instability and rapid aggregation in biological fluids remains problematic. In this study, an injectable hyaluronic acid (HA)-based TRAIL formulation was developed for controlled release of TRAIL while maintaining its stability in vivo. The HA/TRAIL formulations were prepared by simple ionic complexation, and their physicochemical properties were investigated. To confirm formation of TRAIL/HA complex, complex sizes and zeta potential were measured using dynamic laser scattering and the zetasizer. Release tests of TRAIL from HA complex were carried out in vitro using the membrane dialysis method. Bioactivity of the TRAIL/HA complex was evaluated using a HeLa cell line. According to the result, the TRAIL/HA complex formation might be useful as the injectable delivery system of TRAIL for treating tumors.
