Hyaluronic Acid – TRAIL Conjugates for Target Specific Treatment of Liver Cirrhosis |

We previously demonstrated the target-specific delivery of hyaluronic acid (HA) derivatives with an HA modification degree less than ca. 25 mol% to the liver by real-time bioimaging using quantum dots. In this work, HA – tumor necrosis factor related apoptosis inducing ligand (TRAIL) conjugate was developed for target specific systemic treatment of liver cirrhosis. Aldehyde modified HA was synthesized and conjugated with N-terminal amine group of TRAIL. After confirmation of in vitro anti-proliferation activity in HCT116 cells, target specific systemic delivery of HA – TRAIL conjugate was carried out with a pharmacokinetic analysis. The therapeutic effect of HA – TRAIL conjugate on liver cirrhosis was clearly confirmed in animal models. Taken together, the target specific HA – TRAIL conjugate was thought to be successfully exploited for the treatment of liver cirrhosis as a once a week injection dosage form. |
