제목 Preparation and properties of Chitosan-combined PLGA nanoparticle for Oral delivery
초록 The amphipathic PLGA is one of the most successfully used biodegradable polymers because its hydrolysis leads to metabolite monomers, lactic acid and glycolic acid. Because these two monomers are endogenous and easily metabolized by the body, a minimal systemic toxicity is associated with the use of PLGA for drug delivery. PLGA nanoparticle in oral delivery system has many problems, including unstable particle and inefficient absorption in intestinal mucosa. To overcome these obstacles, Chitosan was coated to PLGA nanoparticle, which may lead to enhanced bioavailability. Chitosan-coated PLGA nanoparticle (CPGN) was prepared by w/o/w emulsion method. Its chemical structure was analyzed by 1H-NMR and FT-IR. In addition, its particle size and zeta potential were measured by DLS. Moreover, morphological property of CPGN was observed by TEM. The thermal analysis of CPGN was confirmed by TGA and DSC. Besides, cytotoxicity of CPGN were assessed by MTT assay.
