제목 High-throughput screening of Yarrowia Lipolytica in alginate hydrogel using pneumatic bilayer valve-based microfluidic sorter
초록 Biofuels derived from microalgae showed promisingpotential as a future renewable bioenergy source. Here, we present a microfluidicdroplet sorter (μFADS) for screening randomly mutated Yarroiwa Lipolytica library and sort outthe target subsets exhibiting higher growth rateand increased lipid production First, cell-encapsulated alginate droplets aregenerated within the flow-focusing device, followed by alginate gelationthrough calcium release by lowering pH. The acetic acid in the continuous phasediffused into the droplet due to the difference in solubility, resulting in pHdecrease. Then, the Y. Lipolyticacells were cultured in alginate gels that functioned as an individualmicro-reactor and re-injected into μFADS to evaluate their capacity and sort target cells. As a result, the alginate gelscontaining cells showing 1.5 – 2 times higher growth and lipid production ratewere selectively retrieved compared to the cells in the waste channel.​
