In this research, we prepared liposome containing adenosine triphosphate (ATP), and estimated their surface-enhanced Raman scattering-based assays with Au-Ag alloy-assembled SiO2 nanoparticles. Using the thin film hydration method, ATP as a Raman signal chemical was encapsulated in liposome from a mixture of l-α-phosphatidylcholine, cholesterol, and PE-PEG2000 in chloroform. In the presence of SiO2@Au@Ag, the conditions of ATP for SERS measurement were optimized. The SERS intensity of ATP increased according to the number of ATP-coated liposomes after lysis from 8 × 106 to 8 × 1010. The limit of detection of liposome was 1.3 x 10-17 mol. The results demonstrate the ATP-encapsulated liposome could be applied in the bio-chemical detection with SiO2@Au@Ag NPs. Our approach has a capability to establish a new analytical field for SERS based immunoassay. |
