Aerosol Delivery of Urocanic Acid-Modified Chitosan (UAC)/Programmed Cell Death 4 (PDCD4) Complex for Lung Cancer Therapy |
초록 |
Aerosol gene delivery as non-viral and non-invasive method for gene therapy may provide an alternative for a safer and more effective treatment for lung cancer. In this study, imidazole ring-containing urocanic acid-modified chitosan (UAC) was designed and proven to be efficient for gene delivery in vitro by enhancing the release of the complex into the cytoplasm following the endosomal rupture through proton sponge mechanism. The UAC was used as an aerosol programmed cell death protein 4(PDCD4) gene delivery carrier to apply to the further in vivo study for the lung cancer therapy. Delivered UAC/PDCD4 complex facilitated apoptosis, inhibited cell proliferation and efficiently suppressed tumor angiogenesis. Therefore, our aerosol gene delivery technique is compatible with in vivo gene delivery and can be applied as a non-invasive gene therapy. |
