Fabrication of multi-functional hydrogel based 3D co-culture system for enhancing osteogenesis by paracrine effect |

In this study, paracrine effect between human adipose-derived mesenchymal stem cells (hADSCs) and osteoblasts was systemically investigated in the collagen-based three-dimensional scaffolds for bone regeneration. Osteoblasts-encapsulated microbeads using electrostatic droplet extrusion were embedded with hADSCs in collagen bulk hydrogel scaffolds for indirect co-culture system. We hypothesized that factors secreted from osteoblasts have an influence on osteogenic differentiation of hADSCs by the paracrine effect. We prepared bare and bone morphogenetic protein 2 (BMP-2) containing alginate beads for control groups. This co-culture system in compartmentalized hydrogel based scaffold, osteogenic marker such as ALP, ARS, OCN were increased compared to hADSCs monoculture system. This hydrogel based culture system can potentially be used to study the paracrine effect for various tissue regeneration. |
