In this study, the physical factors for the production of amylase by Arthrobacter sp. were optimized using the reaction surface methodology (RSM). Physical factors including various inoculum volume(0.318-3.682ml), pH(5.318-8.682) and medium volume (16.364-83.363ml) were examined to screening for enhanced amylase production. Amylase activity was as low as 1.66mU/L before optimization after flask culture. Predicted results based on RSM, the optimal condition for distilled production amylase, were 2.84mU/L of estimated amylase production at 2.49mL inoculum volume, pH 6.85 pH and 42.87mL medium volume. Experimental amylase activity was 2.50mU/L, which was increased by 150% as compared with that before the optimization. |
