Improved Pterostilbene Production by Co-culture of E.coli and Streptomyces venezuelae |

Pterostilbene is an effective anti-tumor and antioxidant agent as a resveratrol derivative with two additional methyl branch, thus has significant advantages over resveratrol. The biosynthesis of pterostilbene is derived from phenylpropanoid pathway. The p-coumaric acid is converted to p-coumaric CoA by p-coumarate CoA ligase (4CL) and it combines with three malonyl-CoA to produce resveratrol by stilbene synthase (STS). Next, by series action of 3,5-O-methyltransferase (ROMT), pterostilbene is produced. In this study, 4CL and STS coding genes were transformed into E.coli BL21 (DE3) and ROMT coding gene into Streptomyces venezuelae DHS2001, respectively. Finally, the pterostilbene production was enhanced when the recombinants were incubated in the co-culture system. This result provides the potential utility of the co-culture system for efficient pterostilbene manufacturing. |
