Drug delivering MR imaging agent; DOX@TCL-SPION for in vivo cancer treatment and diagnosis |

We produced a novel drug delivering MR imaging agent, DOX@TCL-SPION for in vivo cancer treatment and diagnosis. We synthesized anti-biofouling polymer coated thermally cross-linked superparamagnetic iron oxide nanoparticles (TCL-SPION) as a MR contrast agent, and then loaded DOX by titration method to prepare stable physical complex. The maximum loading amount of DOX was 2 wt %, and its hydrodynamic size was approximately 21.3 nm. After i.v. injection of DOX@TCL-SPION to LLC allograft mice, a noticeable darkening appeared in the tumor area in the T2 weighted MR imaging with a RSE value of about 58 %. After sacrifice mice, ex vivo organs in IVIS images indicated exclusively intense fluorescence signal of the DOX in the tumor. Consistent with this, DOX@TCL-SPION (0.64 dox eq.mg/kg) exhibited superior therapeutic outcome, when compared with free DOX (0.64 mg/kg or 5 mg/kg). Therefore, we anticipate that DOX@TCL-SPION may be utilized to develop novel therapeutic and diagnostic modalities. |
