Although 5-fluorouracil (5-FU) is one of the major antimetabolites widely used in a variety of solid cancers as antitumor agents, it also has undesirable side effects such as high incidences of toxicity in the bone marrow, gastrointestinal tract, central nervous system and skin. In comparison with the low molecular weight drug, a polymer-drug conjugate can be expected to overcome the problem of side effects mentioned above and to have a prolonged duration of activity. In this study, a methacrylamide derivative of 4-aminosalicylic acid monomer was synthesized, characterized and polymerized directly via radical polymerization followed by the conjugation reaction with 5-fluorouracil by ester linkage. The synthesized polymers were characterized by FT-IR, 1H-NMR spectroscopies and gel permeation chromatography. |
