제목 Combination of hot spot mutations of antibody Fc for enhanced therapeutic effector functions.
초록 The Fc region of IgG antibody plays a pivotal role in eliciting various immune effector functions by the interaction between antibody and its various ligands such as FcγRs. To isolate Fc variants exhibiting improved immune cell mediated-therapeutic effector functions, we have constructed a library composed of Fc variants containing the combination of the mutations which were previously identified for enhanced FcγR binding. Using a bacterial display in coupled with a flow cytometry-based high throughput library screening, we have isolated Fc variants which allow remarkably improved FcγR binding affinity. The screening strategies, binding characteristics of the isolated Fc variants, and potential applications will be discussed.
