Polysuccinimide (PSI) were synthesized as an intracellular drug carriers. Hydrophobic octadecylamine (C18) and hydrophilic O-(2-aminoethyl) polyethylene glycol (PEG) were grafted on a PSI backbone for amphiphilicity to make micelles structure. Folic acid was used as targeted ligand and conjugated to PEG/ Hyd-Curcumin/C18-g-PSI polymer to facilitate active targeting to colon cancer cells. Two different formulations: Folate-PEG/Hyd-Curcumin/C18-g-PSI and PEG/Hyd-Curcumin/C18-g-PSI were employed. Comparatives studies on the cytotoxicity and modulation Wnt/beta-catenin signaling pathway of the different formulations, in the presence and absence of acid folic, were carried out by MTT assay and TOP Flash assay, respectively. Finally, the active targeting of the polymeric micelles to folate-expressing cells was confirmed upon in SW480 colon cancer cells and suppression beta-catenin to prevent from acting targeted gene. |
