ZVI (Zero-Valent Iron) acts as an electron donor to provide reducing equivalent for 1,3-PDO synthesis from glycerol. The electron from ZVI oxidation activates the reductive pathway, causing changes of metabolite productions. In this study, the effect of ZVI on the metabolic shift of Klebsiella pneumoniae L17 which has been known as electroactive species. The NADH ratio was significantly increased with ZVI as compared to that without ZVI (21: 6). To quantitatively measure the expression level of a specific protein with qPCR, we designed primer for dhaB, dhaD, glpK and rpoD genes. The increase of glycerol dehydratase (DhaB) was confirmed in ZVI. Therefore, When ZVI was supplemented, the reductive pathway was activated and 1,3-PDO production was increased. These results indicate that ZVI modulate the biotransformation of electroactive strain such as K. pneumoniae L17, and enhance the conversion of glycerol into a value-added platform chemical. |
