Biomimetic PLGA/ECM 3D porous scaffolds for kidney tissue regeneration |

Chronic kidney disease is now recognized as a major health problem, but current therapies have many limitations. Consequently, biodegradable scaffolds to help repairing injured tissue are emerging as a promising approach in the field of kidney tissue engineering. Poly(lactic-co-glycolic acid) (PLGA) is a useful biomedical material, but its insufficient biocompatibility impeded renal regeneration. In this work, we developed extracellular matrix (ECM) incorporated PLGA (PLGA/ECM) porous scaffolds as a cell supporting material. PLGA/ECM scaffolds were prepared and characterized through various analyses. Cell proliferation on the scaffolds increased with an increase in ECM concentrations. The PLGA/ECM scaffold was shown to be an effective matrix for the reconstitution of glomerulus in partially nephrectomized mice, compared with only PLGA control. These results suggest that the ECM incorporated PLGA 3D porous scaffolds could be one of the promising materials for kidney tissue regeneration. |
