Heparin immobilization has been widely studied and clinically used for development of antithrombogenic implants such as vascular grafts. Additionally, cell adhesive peptides can be applied for an improved bloodcompatibility through the induction of endothelialization. In this study, bioactive PU surface which can be shown the antithrombogenicity and promote endothelialization was developed by using heparin, YIGSR, and RGD. Heparin and peptides immobilized PU surface was sequentially prepared. After grafting of OCN-PEG-NCO spacer group through an allophanate reaction with PU and then heparin and peptides were covalently bound to the remaining isocyanate group. These surface-modified PU was characterized by contact angle, Toluidine blue assay and Amino acid analysis. We have also demonstrated significant retention of heparin bioactivity with anti-factor Xa. These studies represented the PU was successfully immobilized with bioactive molecules for anticoagulation and endothelialization. |
