제목 Tissue Engineered Biodisk using Natural/Synthetic Mesohybrid Scaffolds
초록 Study on tissue engineered biodisk as the substitute therapy for the degeneration of the intervertebral disc composed of two distinct regions, the inner soft and highly hydrated nucleus pulposus and the outer firm annulus fibrosus is active. Actually the regeneration of nucleus pulposus on the upper surface of a porous bone substitute material (Calcium polyphosphate (CPP)) was performed by Rita A. Kandel et. al. We tried to prepare Biodisk based on mesohybrid scaffolds included a new bone formation material, DBP. Demineralized bone particle (DBP) impregnated with bone morphogenetic proteins (BMP) takes effect as the powerful inducer of a new bone growth. It has been reported that the above-stated BMP expedites the differentiation of bone marrow stromal cells (BMSCs) into bone and cartilage. Poly(lactide-co-glycolide) (PLGA), one of biodegradable polymers, has been used widely for tissue engineering, recently.
Mesohybrid scaffolds created with natural/synthetic materials by solvent casting/salt leaching method is 5mm diameter and 5mm height. In order to analysis the surface and the inner structure scaffolds were observed by means of SEM and the average pore size of them was 250㎛.
In this study cells collected directly from disks of distal caudal spines were used. Cells were seeded on scaffolds in the specified medium according to the known procedure and then harvested at 2, 4, 6 weeks to analyze the histologic appearance, the water content, proteogylcan and collagen contents, etc.
