제목 | Preparation and Characterization of the 1,25(OH)2 Vitamin D3-loaded Poly(lactide-co-glycolide) Scaffolds for Tissue Engineered Bone |
초록 | 1,25(OH)2 vitamin D3-loaded biodegradable PLGA scaffolds are synthetic bone substitutes that promote bone formation by osteoconduction and osteoinduction. 1,25(OH)2 vitamin D3 plays as a regulator of osteogenic differentiation of bone marrow stromal cell (BMSC). These scaffolds were confirmed in vivo osteogenic potential for tissue engineered bone. Biodegradable scaffolds were fabricated by salt leaching/solvent casting method. Poly(lactide-co-glycolide) (PLGA 75:25, Mw 90000 g/mole) was dissolved into methylene chloride and this solution was mixed with sodium chloride and 1,25(OH)2 vitamin D3. molded, and then leached in an aqueous solution. The scaffolds had relatively homogeneous pore structures throughout the matrix and showed an average pore size in the range 150~280 μm and over 90% porosity. From the results, this sustained, besides, constant localized release system can potentially provide 1,25(OH)2 vitamin D3 and encourage calcium and phosphate, which plays an important role for osteoconduction and osteoinduction. These scaffolds may be used for the applications of bone regeneration. In vivo and detailed studies on the 1,25(OH)2 vitamin D3-loaded scaffolds are now in progress.
