Cancer chemotherapy has become increasingly utilized, but there are limitations against disseminated cancer because of resistance treatment with traditional anticancer drug. The major mechanism of drug resistance was the expression of an energy dependent drug efflux pump, such as P-glycoprotein (P-gp). Most anticancer drugs are substrates for this transport system by the substrate-binding region of P-gp, decreasing cellular uptake. To overcome drug resistance, we developed polymeric micelle conjugated photosensitizers (PMCP), which is based on nonionic triblock copolymer conjugated photosensitizers. The doxorubicin-loaded PMCP are collapsed via lipid peroxidation with singlet oxygen through photosensitizers. Photochemical internalization (PCI) is a light triggered strategy to breakdown of the cellular membrane. Further, we have expected for this study to represent a potential platform for drug resistant cancer cell therapy. |
