제목 A fluorescence-based miRNA detecting probe in cancer cell using Polyvalent DNA-modified gold nanoparticle
초록 A sensitive and specific detection method is urgently needed for intensive studies on different expression and regulatory role of microRNAs involved in the initiation and progression of cancer. Here, we present a simple and reliable method for the quantification of microRNA using polyvalent molecular beacon functionalized gold nanoparticles (MB-AuNP). MBs are comprised of stable duplex of anti-miR strand complementary to the target miR and fluorophore-labeled short strand. In this conformation, fluorophore to the AuNP surface leads to quenching. Also gold nanoparticles densely packed with MB are less susceptible to degradation by DNase and have greater cellular uptake efficiency than each MB. When a target miRNA is bound to the complementary anti-miR strand, fluorophore-labeled strand is becoming fluorescent. MiRNA detection and measurement miRNAs level within live cells research through MB-AuNPs would be contributed our fundamental understanding for cancer research and treatment.
