Development of caprolactam-specific RNA based biosensor |

Caprolactam is a monomer of nylon-6, a high-tensile material of synthetic fiber. An intracellular biosensor for caprolactam can facilitate high-throughput metabolic evolution of microbial strains. Because of the mixed production of caprolactam and valerolactam in the recombinant strain, a biosensor should specifically recognize the structure of caprolactam. However, a highly specific caprolactam biosensor has yet been reported. Here, we developed an artificial riboswitch that meets the need for sensor-guided evolution. Coupled in vitro-in vivo library selection strategy was applied for the development of the translation-initiation regulating sensor. Selected caprolactam riboswitch successfully discriminated caprolactam from valerolactam and enabled caprolactam-dependent control of cell growth. Further applications in microbial cell factory for caprolactam production using caprolactam biosensor is expected. |
