Targeted gene delivery of the cationic lipid emulsion system containing folate-PEG-DPPE |

Gene therapy depends on the efficient delivery of genetic material into the nucleus and its effective expression within the cell. Cationic lipid system has been widely used in pharmaceutical and medical fields as used as drug and gene carriers. DOTAP(1,2-Dioleoyl-3-Trimethylammonium-Propane) was used as a cationic lipid. We conjugated the tumor-targeted ligand to the polymeric lipid. Folate was conjugated on PEG-DPPE-amine. The mean particle sizes of polymeric lipid system and DNA/lipid complex system were 206.6nm and 150.5 nm, respectively. We tried to check the targeting effect of folate ligand of polymeric lipid system. HeLa and 293 cell was used for the in Vitro transfection experiment. HeLa cell is a folate positive cell line. The transfection efficiencies was 100 times higher than that of DOTAP emulsion due to the targeting effect of folate. |
