For the last decades, cell sheet engineering has emerged as a new paradigm for effective delivery of therapeutic cells in tissue engineering. The harvested cell sheet can maintain cell-extracellular matrix and cell-cell interactions. In such a process, development of cell-culture substrate to harvest single cells or cell sheet without enzyme treatment is a critical step. Various studies have been progressed to harvest cell sheet with controlled cell detachment capacity. However, current methods had some limitations. In this study, we investigated the effect of hydrogel fabrication conditions on various characteristic. First, we examined the effect of polymer concentration on gelation time of hydrogels and thermo-sensitive characteristics of hydrogel. The difference of cell adhesion and attachment morphology on various hydrogels with varied mechanical properties was then studied, and we indirectly confirmed cell detachment mechanism using target substrate with different characteristics. |
