In this study, we designed a gene carrier to therapy the disease related with cardiovascular, such as atherosclerosis, restenosis. A specific cell targeting peptide, artery wall binding peptide (AWBP), was conjugate to poly(ethylene glycol)-grafted-poly(L-lysine) (PEG-g-PLL) to enhance the gene transfer to artery wall cells. It is well known that PLL interact hydrophobically with low density lipoprotein (LDL) and AWBP-PEG-PLL deliver the gene by receptor-mediated endocytosis process. Also, PEG was introduced for long circulation of carrier in the blood. To identify the synthesis of AWBP-PEG-PLL, 1H-NMR was measured. The particle size and the shape of AWBP-PEG-PLL/pDNA complexes were measured by dynamic light scattering (DLS) and atomic force microscopy (AFM). To show the potential as a gene carrier, we performed the agarose gel retardation assay and transfection efficiency in both bovine aorta endothelial cell and smooth muscle cells. After this study, it will be continuously investigated for the synthesis of gene carrier using polyethylenimine (PEI) and low molecular weight water soluble chitosan (LMWSC).
