제목 Injectable mesoporous silica microparticles enabling effective recruitment and activation of dendritic cells for anti-cancer immunotherapy
초록 Cancer immunotherapy has intensively studied for decades including adaptive cell therapy and immune checkpoint blockade. Dendritic cells (DCs) are representative antigen-presenting cells to deliver antigenic information to the adaptive immunity. Therefore, recruiting and educating DCs are one of the main purposes in immunotherapy. To develop effective in situ immune cell recruitment, we applied two kinds of mesoporous silica particles; (1) mesoporous silica rod (MSRs), (2) mesoporous cellular foam (MCFs). In this study, we investigate how the size and morphology of mesoporous silica microparticles can affect the recruitment yield of host immune cells and antigen delivery pathway. After subcutaneously injection into mice for 7 days, both MSRs (90 um length with hexagonal pore) and MCFs (6.7 um size with inter-connected pore) form the nodule while MCFs recruit higher number of DCs. MCFs can activate immature DCs by acting as DC-recruiting scaffold via chemokine release and carrier to deliver antigen into cytoplasm of DCs. These feature of MCFs led to an effective inhibition of tumor growth in prophylactic and lung metastatic cancer model.
