Regulation of SDF-1 induced MSC migration by MSC-EC interaction on Biomimetic microfluidic gradient chip |

Stem cell homing is a crucial process in stem cell biology as well as in cell therapy. The mechanisms of directional migration of stem cells by homing factors is not well known although extensive researches are done by scientists. In this study, we used the 3-channel- microfluidic device to mimic microenvironments of stem cell homing and to study SDF-1 induced directional migration of mesenchymal stem cell (MSC) interacting with endothelial cell (EC). The microchip consists of EC monolayer in the middle channel and the MSC suspension media flows through the endothelium-coated channel by micropumps. We quantified the kinematic analysis of stem cell migration through endothelium and hydrogel scaffold. Moreover, we attempted to control cell migration by inhibiting MSC-EC interaction with VLA-1 and LFA-1, and stem cell homing with CXCR4 and Rho/ROCK. Ultimately, this in vitro quantitative system will be able to visualize in vivo and be used as a drug testing platform. |
