Injectable cell-cross-linked gels for tissue engineering |

Alginate has been widely used for tissue engineering due to biocompatibility, simple gelation behavior, and ease of administration into the body in a minimally invasive manner. We hypothesized that mechanical strength and gelation time of cell-cross-linked gels can be regulated by addition of minimal amount of ionic cross-linking molecules, as well as by variation of alginate composition. An RGD-modified alginate solution, mixed with minimal amount of calcium sulfate, did not show significant changes in the viscoelastic properties. However, G' of the solution was substantially increased when mixed with NIH 3T3 cells, indicating the formation of stable and strong cell-cross-linked gels. The addition of small amount of divalent cations was able to enhance the physical properties of cell-cross-linked gels and reduce gelation time, depending on the molecular composition of alginate backbone. |
