Antimicrobial peptides(AMPs) make up the primary defense mechanism in many organisms, and have broad target spectrum. AMPs have attracted attention as a new class of antibiotic substances. Research of many AMPs is actively under way to debelop engineered AMPs having increased activity. However, AMPs are difficult to express in live host cells because of their toxicity In this study, we examined to use cell-free translation as an altermative platform for the expression of AMPs. Cell-free translation does not depend on cell viability and is highly flexible in changing the expression conditions and components. At the same time, E.coli cells were palced in the same reaction mixture expressiong pesoganan After the cell-free translation reaction was completed, viable cells in the reaction mixture was counted. As a result, we were able to accomplish simultaneous expression and assessment of the AMP without seperate purification steps. |
