Chitosan as a biocompatible natural polysaccharide has been investigated as versatile polymeric scaffolds for the systemic delivery platforms and locally injectable gels. However, many of such chitosan derivatives often suffer from low aqueous solubility over a wide range of pH conditions and extremely high viscosity, which significantly limits their broad applicability. We present here chemically modified chitosan derivatives whose conformation can be modulated to overcome such limitations. In the first approach, chelation of heavy metal cation can induce the conformation change of chitosan, allowing for the viscosity modulation. Secondly, pyrene modification further enables the fluorescence monitoring of conformation change via excimer formation. Their characterization and colloidal properties will be discussed. |
