The statistical associating fluid theory (SAFT) equation of state is employed to represent the protein-protein interaction in the two-protein systems. The equation of state is presented as sum of three Helmholtz energy terms: segment term(temperature-dependent hard sphere), chain term(increment due to chain formation), and association term(increment due to association). This equation of state has been developed by extending Wertheim’s theory obtained from a resumed cluster expansion. To describe properties of protein systems, shape factor and pre-aggregation factor is introduced for corresponding protein/salt systems. Those factors are then introduced to calculate phase behavior in the aqueous two-protein system. Agreement with experimental data is reasonable in the given pH and ionic strength range with no additional binary parameters. |
