As an alternative of plant-derived triacylglycerol or animal fat, free fatty acid produced by microorganism provides promising source to synthesize oleochmicals. Recombinant E. coli strains with deletion of envR, gusC, or mdlA, introduction of an acetyl-CoA carboxylation bypass, and expression of an acyl-CoA thioesterase I mutant with high specific activity exhibited several fold increase in the total FFA production, respectively. The production of DCAs by the whole-cell biocatalysis is often limited by cofactor regeneration. Here, the ω-oxidation pathway genes were co-expressed with xylose reductase (XR) gene to regenerate cofactors in an engineered E. coli strain that co-metabolizes glucose and xylose. The resulting strain enhanced the production of DCAs by 344% over the control system without expressing XR and produced xylitol in the presence of xylose. |
