Incorporation of Simvastatin loaded PLGA microcarrier in a heparin mimicking polymer conjugated with BMP-2 as dual drug delivery system for bone tissue regeneration |

BMP-2 like other bone morphogenetic proteins plays an important role in the development of bone and cartilage. It is involved in the hedgehog pathway, TGF beta signaling pathway and in cytokine-cytokine receptor interaction. Simvastatin has been reported to promote osteoblastic activity and inhibit osteoclastic activity. It is also reported to have an anti-inflammatory effect that works by decreasing the production of interleukin-6 and interleukin-8. But BMP-2 is hydrophilic whereas Simvastatin is hydrophobic. The main challenge of this work is to incorporate both type of bone regeneration factor in single system to elevate the system efficacy. Controlled and sustained release profile should take in consideration to design the system. Regarding all the factors Simvastatin was incorporated in PLGA micro carrier whereas BMP-2 was stabilized by conjugating with poly(sodium 4-styrenesulfonate-co-poly(ethylene glycol) methyl ether methacrylate) (p(SS-co-PEGMA)). To evaluate the system physically SEM imaging, FT-IR, gelation time and degradation checking were undertaken. MTT assay and Confocal imaging with pre-osteoblat cells were the part of biological evaluation or cell viability of this novel complex system. Evaluation of osteogensis of the system was very important which was examined through RT-PCR. Finally In-vitro release profile was checked for both Simvastatin and BMP-2 which patented controlled, sustained and prolonged release in each case. |
