HAp Granules Incorporated in Injectable Bone Substitute System for Improved Mechanical Strength and Its In-Vitro Evaluation for Bone Tissue Regeneration |

To meet the demand of bone regeneration due to fracture or disease, there are many reports on the fabrication of injectable bone substitutes which are combined with bio-ceramics and bio-polymer self hardening injectable bone substitute as a common approach. In this work, we fabricated spherical HAp granules to incorporate in IBS (injectable bone substitute system) for the improvement of strength of set IBS and to be used as a drug delivery system. PVA-Gel-HAp composite slurry was sprayed by sprayer on liquid nitrogen maintaining the flow rate, distance of spraying, high pressure and kept into -80 degree freeze and then freeze-drier to make granules. After the lyophilization, the granules are kept into microwave sintering machine for sintering at 13500 C in order to burn out polymer and make porous HAp granules. Then HAp granules were incorporated with Injectable Bone Substitute system for higher compressive strength. Detailed morphological and material characterizations such as compressive strength, porosity, microstructure analysis were carried out. MTT assay, confocal imaging were carried out as in vitro experiments to observe the cell attachment, proliferation with pre-osteoblast MC3T3-E1 to evaluate the IBS.. |
