Biosynthesis of Spectinomycin: A Tool for the Production of Hybrid Antibiotics |

Spectinomycin, a broad-spectrum aminoglycoside antibiotic, is produced by S. spectabilis. It has a unique structure in which a sugar component actinospectose is linked with diaminocyclitol moiety of actinamine by β-glycosidic and hemiketal bonds. Out of the several ORFs found to be involved in the biosynthesis of Spectinomycin, we have cloned and characterized some crucial genes like dTDP glucose synthase, 4,6-dhydratase, myo-inositol dehydrogenase, scyllo-inosose aminotransferase and N-methyltransferase by heterologous gene expression in E.coli. Similarly, the function of N-methyltransferase gene is identified by gene inactivation experiment resulting a novel antibiotic i.e. non-methylated spectinomycin. To facilitate the characterization of these biosynthetic genes, a new vector (pIBR100) has been constructed that can be used for the high level expression in actinomycetes. With the help of pIBR100, we are developing a gene cassette for the overproduction of actinamine that can be conjugated to various aglycones to pave the way for the biosynthesis of novel and high potential bioactive materials. |
