제목 Biosensing of cardiovascular disease biomarkers by engineering immune cells with aptamers specific to the target molecules
초록 Aptamers can be easily modified by changing its sequences. Their usage in place of antibodies due to their binding affinities and stability. Aptamer can be used as a detecting probe or linker.
CRP is a protein which indicates the progress and prognosis of inflammatory diseases. CRP is one of a well-known biomarker for acute inflammatory events; it is therefore necessary to measure CRP levels in the blood accurately.
We demonstrated the preparation of single-stand DNA aptamer-conjugated with live immune cell; peripheral blood mononuclear cells (Apt-PBMCs) that specifically capture human CRP. Capture ability of Apt-PBMCs against CRP were tested. Additionally, cytokine release profiling assay, cell viability test against conjugation steps or buffers, and the migratory behavior toward highly concentrated CRP sites were tested.  
In summary, engineered Apt-PBMCs have potential applications as live cell biosensors and for in vivo and in vitro tracing of CRP secretion sites.
