Electrochemical Label-Free Sensor for Detection of Botulinum Neurotoxin Type E Light Chain |

The highly sensitive sensor has required for the detection of Botulinum neurotoxin (BoNT), since it has the potential to be used as the biological weapon due to their toxicity with the high fatality in a low dosage. In this study, we developed the simple label-free electrochemical sensor for the detection of BoNT type E light chain (BoNT/E LC). In the dual mode cyclic voltammetry system, the redox signal of ferri-ferrocyanide was decreased due to the steric interruption of peptides designed to be cleaved by BoNT/E LC. After the introduction of BoNT/E LC, the redox signal was recovered because the peptides on the gold electrodes are cleaved by enzymatic reaction. Through this method, it was able to detect BoNT/E LC with a concentration as low as 5 pg/ml. We expected that it is possible to improve the performance of the sensor by modifying the geometry of the device. |
