Absorption through the intestinal epithelium is a major barrier through which orally administered drugs must pass. Therefore, intestinal absorption research is important in the field of drug development. However, animal experiments or human clinical trials, which are conventional intestinal absorption models, are limited in terms of ethical issues and economics. As an alternative to the conventional intestinal absorption model, an organ-on-a-chip was developed. In this study, we developed a multi-organ chip that organically connects the gut-liver-kidney. We confirmed that cell culture on the chip was possible by culturing human intestinal epithelial cells(Caco-2 and primary cells) on the chip. And we confirmed the intestinal function of the cultured enterocytes on the chip. Also, by checking the intestinal permeability of the actual drug, it was confirmed that it is possible to observe the intestinal absorption in the chip. Furthermore, by comparing the cell line Caco-2 with primary cells, more suitable cells could be selected as a model for intestinal absorption. We hope that the chip could potentially be used as a new and more accurate intestinal apsorption model. |
