제목 | Tyrosinase-catalyzed hyaluronic acid hydrogel and its application in meniscus engineering |
초록 | Meniscal tissues have limited regenerative capacity, and the treatment for meniscal defects has been limited to arthroscopic meniscectomy or meniscal suture. In recent years, intra-articular injection of hydrogels is emerging as a promising alternative to conventional therapy, with growing demand of minimally invasive treatments [1, 2]. Herein, we developed an enzymatically crosslinking hydrogel which can be introduced into the knee joint as an injectable treatment of damaged meniscus tissue. The hydrogel was based on hyaluronic acid (HA), which is found in high concentration in the synovial fluid and functions as a lubricant in the joint. Tyramine-conjugated HA/gelatin hydrogel was formed via oxidative coupling mediated by tyrosinase. The crosslinking density and degradation kinetics of hydrogels were tuned by controlling the degree of tyramine substitution and the concentration of tyrosinase. Tyrosinase-catalyzed hydrogel showed strong biocompatibility for meniscus cells, compared to other crosslinking enzymes such as horseraedish peroxidase. Therefore, we suggest that tyrosinase-catalyzed HA/gelatin hydrogel as a potential applicable biomaterials for meniscus tissue engineering. [1] H. Pereira, et al.
