Exosomes are nano-sized vesicles and detected in body fluids. They contain useful biomarkers such as miRNAs produced from their originating cell. This makes exosome miRNAs from the body fluids an attractive biomarker in disease diagnosis. However, current techniques, including real-time PCR analysis, are time-consuming and laborious, making them unsuitable for exosome miRNA detection for diagnosis. Herein, in situ single step detection of multiple miRNAs in a whole exosome has been developed as a novel diagnosis method that can be utilized for various diseases. Nano-sized oligonucleotide probe, molecular beacon (MB) was constructed for the detection of exosomal miRNAs. The method described here is simple, fast, and sensitive, so it will offer great opportunities for the high-throughput diagnosis and prognosis of diseases. |
