제목 Bioactivatable Poly(ethylene glycol)-Doxorubicin Conjugate for Targeted Cancer Therapy
초록 In this study, we synthesized a bioreducible methoxy poly(ethylene glycol)-doxorubicin (mPEG-ss-DOX) conjugate for targeted cancer therapy. Anticancer agent DOX was chemically conjugated with mPEG via a bioreducible disulfide bond. The amphiphilic mPEG-ss-DOX self-assembled in aqueous media to form nanoparticles (NPs) and efficiently encapsulated superparamagnetic iron oxide nanoparticles (SPIONs) to form the SPION-loaded NPs. The SPION-loaded NPs showed a narrow size distribution with an average size of about 80 nm and demonstrated a good colloidal stability. The bioreducible dissociation and drug release properties of the NPs were observed in the presence of dithiothreitol (DTT). The iron oxide could be encapsulated up to ~60 wt% proportionally to the feed ratio. The SPION-loaded NPs showed not only lower toxicity than free DOX, also the tumor targeting and imaging potential. The resulting SPION-loaded NPs could be useful for cancer MR imaging and chemotherapy.
