제목 Drug delivery of felodipine using multi-coating beads
초록 Hypertension is a common disorder in current societies and associated with an increased cardiovascular risk. Felodipine (FD), a vasoselective calcium channel antagonist of the dihydropyridine class, is widely used as a potent antihypertensive drug. We designed some controlled release of devices FD for improvement of patient compliance. We prepared a few kinds of FD beads were composed of sugar, various excipients, such as hydroxypropyl methyl cellulose, and eudragit using fluid bed coater. The dissolution test was carried by the paddle method # 2 at a stirring speed of 50 rpm at 37℃ in simulated intestinal fluid with tween 80 (w/v%). Drug contents and release amounts of FD were analyzed by high performed liquid chromatography (HPLC). Based on the results of release test, FD granules release rate was associated with composition of eudragit membrane, the desired ratio of membrane, such as eudragit RS/RL ratio, 9:1 and 8:2, and the amount of component of beads, such as plasticizer and disintegrant. Our devices were sustained release in 8 hours and release profiles were seemed to the near zero-order release.
